Ondansetron (Zofran) – Side Effects, Uses, Mechanism of Action, Dosage, Interactions, Warnings
Ondansetron, marketed under brand names such as Zofran, Zofran ODT, and Zuplenz, is a medication used to prevent nausea and vomiting associated with cancer chemotherapy, radiation therapy, and surgery. It functions as a serotonin 5-HT3 receptor antagonist, blocking serotonin's action in the chemoreceptor trigger zone and on vagal nerve terminals in the gastrointestinal tract. Common side effects include headache, dizziness, drowsiness, constipation, and fatigue, while serious side effects may include QT prolongation, serotonin syndrome, and allergic reactions. Dosages vary based on the condition: for chemotherapy-induced nausea, adults typically take 8 mg twice daily or 24 mg as a single dose, and for radiation-induced nausea, 8 mg orally before radiotherapy. Ondansetron interacts with drugs that prolong the QT interval, serotonergic drugs, apomorphine, and CYP3A4 substrates. Warnings include risks of QT prolongation, serotonin syndrome, hypersensitivity reactions, and caution during pregnancy and breastfeeding.
