Triptans, which include the archetypal drug sumatriptan, are a class of drugs used to treat headaches. They act as agonists or activators of Serotonin receptors, specifically the serotonin 1B and 1D receptors in the trigeminal nerve and blood vessels in the brain. This agonist activity inhibits the release and delivery of vasoactive peptides that cause inflammation associated with headache disorders. Specifically, triptans are used in the acute treatment of migraine and cluster headaches. An important side effect of taking triptans is the development of coronary vasospasms. As such, these drugs are contraindicated in patients with a history of coronary artery disease or Prinzmetal angina. For those who understand the debilitating effects of a migraine headache, knowing relief may be a spray away in 15 minutes could be a game changer.
"Given that it's an intranasal, it has fast onset, which is helpful for patients who have migraine peak very quickly," Dr. Emad Estemalik, headache specialist at Cleveland Clinic, said.
The FDA approved Pfizer's Zavzpret on Friday, and the drug is expected to have an official launch in July. We'll find out then how much it will cost, although Pfizer says it will be comparable to other migraine medications.
The spray is part of a class of drugs that stop migraines when they start.
"For a lot of patients who have significant nausea or vomiting, this is an ideal treatment because they don't have to take anything oral in terms of medication; just use an intranasal medication to quickly abort their migraines," Estemalik explained.
Zavzpret is approved for those with or without migraine auras. Studies showed the medication could give pain relief in 15 minutes, normal function in 30, and a return to normal in two hours. However, the label has a warning to stop taking it in case of facial swelling and skin rashes.

Relpax is a medicine used to relieve migraines.
The active ingredient in Relpax is eletriptan, which is a type of substance called a serotonin agonist. Eletriptan is also more commonly known as a triptan.
Although the cause of migraine attacks is not fully understood, it is thought that widening of blood vessels in the brain causes the throbbing pain of migraine headaches. Eletriptan relieves this pain by causing the blood vessels in the brain to narrow.
Eletriptan works by stimulating receptors called serotonin receptors that are found in the brain. A natural substance called serotonin normally acts on these receptors, causing blood vessels in the brain to narrow. Eletriptan mimics this action of serotonin by directly stimulating the serotonin receptors in the brain. This narrows the blood vessels and so relieves the pain of migraine headaches.

What Is Reyvow Used For?
It's a newer (at the time of this video) medication you take by mouth to treat acute migraine attacks.
How Does It Work In Your Body?
Is stimulates a receptor in your body that helps 'calm' the trigeminal system - which is a part of your brain.
It's main advantage is that it really focuses on lowering the pain associated with migraine without causing vasoconstriction ... which is what the triptans do.
Another unique thing about Reyvow is that it acts not just peripherally (like all the other acute migraine medications we have) - but also centrally.
How Is It Dosed?
It's availabe as a 50, 100, or 200 mg dose. But unlike other acute migraine meds you can only take 1 dose in 24 hours.
And the safety of treating more than 4 migraine attacks in a month has not been established.
Is It A Triptan?
The classic class of drugs used to treat migraine attacks have been around since the 1990's and are called the 'triptans'.
But triptans act in a different way than Reyvow.
Side Effects of Reyvow?
Reyvow doesn't have a lot of side effects but the one it can cause is dizziness in up to 9-17% of users.
But the dizziness can be severe enough that you CANNOT USE REYVOW IF YOU NEED TO DRIVE A CAR OF MACHINERY WITHIN 8 HOURS OF TAKING IT!
How Fast Does It Work?
You can start to notice improvement of your migraine within 30-60 minutes of taking Reyvow.

Imigran contains the active ingredient sumatriptan, which is a type of medicine called a serotonin agonist. This type of medicine is also commonly known as a triptan. It is a painkiller specifically used to relieve migraine attacks.
Although the cause of migraine attacks is not fully understood, it is thought that widening of blood vessels in the brain causes the throbbing pain of migraine headaches. Sumatriptan relieves this pain by causing the blood vessels in the brain to narrow.
Sumatriptan works by mimicking the action of serotonin by directly stimulating the serotonin receptors in the brain. This narrows the blood vessels and so relieves the pain of migraine headaches.

Sumatriptan is used to manage migraine headaches and it causes cranial vasoconstriction.
It is available as tablets and it can be used at a dose range of 25 to 100 mg. It can also cause coronary vasoconstriction that induces chest pain, jaw pain and arrhythmias. It is 5-HT 1B/1D receptor agonist. Rarely it can cause vision changes.

Abstract: Subcutaneous sumatriptan is an effective treatment for pain from acute migraine headache, and can be used in patients with known migraine syndrome and in patients with primary headaches when secondary causes have been excluded. In limited comparative trials, subcutaneous sumatriptan performed in a manner comparable with oral eletriptan and intravenous metoclopramide, was superior to intravenous aspirin and intramuscular trimethobenzamide-diphenhydramine, and was inferior to intravenous prochlorperazine for pain relief. The most common side effects seen with subcutaneous sumatriptan are injection site reactions and triptan sensations. As with all triptans, there is a risk of rare cardiovascular events with subcutaneous sumatriptan and its use should be limited to those without known cerebrovascular disease and limited in those with known cardiovascular risk factors and unknown disease status. In studies of patient preference and tolerability, the subcutaneous formulation has a faster time of onset and high rate of efficacy when compared with the oral formulation, but the oral formulation appears to be better tolerated. It is important to consider the needs of the patient, their past medical history, and what aspects of migraine treatment are most important to the patient when considering treatment of acute migraine or primary headache. Subcutaneous sumatriptan is a good first-line agent for the treatment of pain from acute migraine headaches and primary headaches.

A migraine is a medical disorder characterised by recurring headaches that are moderate to severe. Typically, a migraine headache will present as a throbbing pain on one side of the head, but it can often be both.
Unlike typical headaches, migraines often present with additional symptoms that can interfere with daily life. These symptoms can either appear before, during or after the pain phase of a migraine.
If you’re struggling with migraines, it’s important to remember that you’re not alone. Migraines are common in the UK, with as many as one in five women and one in 15 men suffering from the condition. Migraines will usually begin in early adulthood, and vary significantly in frequency from person to person. One migraine sufferer might experience an attack every six months; another might get one every few days.
Triptans are a family of medications used to treat migraines that do not respond to ordinary painkillers and rest. These treatments are said to work by binding to serotonin receptors in the brain’s blood vessels and nerve endings. This is then believed to block the release of inflammatory neuropeptides, which are protein-like molecules that neurons use to communicate with each other.
