Upon phosphorylation by the protein tyrosine kinase from activated T cells after organ transplantation, hydrogelators in the tacrolimus‐coassembled nanofibers will be efficiently converted to their hydrophilic phosphates, resulting in the disassembly of the nanofibers and the release of tacrolimus for T‐cell inhibition. This is reported by Jindao Wu, Zhen Zheng, Yuanyuan Chong, Xiangcheng Li, Liyong Pu, Qiyun Tang, Liu Yang, Xuehao Wang, Fuqiang Wang, and Gaolin Liang in the article https://doi.org/10.1002/adma.201805018. Tacrolimus.

Calcineurin inhibitors such as tacrolimus and cyclosporine are immunosuppressants. They are used mainly to prevent organ transplant rejection in combination with mycophenolate and steroids as well used in severe atopic dermatitis. They are not really used as much for rheumatologic conditions any more, possibly for lupus nephritis. Calcineurin inhibitors are also used for the treatment of graft versus host disease in bone marrow transplant recipients. Calcineurin inhibitors inhibit the synthesis of IL2.

Any medication have downsides, even if it's demonstrated an excellent safety record. Let's talk about possible side effects from using Protopic ointment and Elidel cream.
UPDATE: in summer 2024, my skin stopped reacting to sun! I guess that this side-effect was caused solely by Protopic, which accumulated in my organism for years of use, and Elidel hadn't replaced it yet it in summer 2023, when I experienced severe sun allergy.

“I feel lousy should I blame Tacrolimus/ Prograf?” The viewer went on the add “I feel fatigued, upset stomach, headaches, overall drained.” Depending on your symptoms probably yes, Tacrolimus Or Prograf Can Make You Feel Lousy. Even though Tacrolimus is one of the most important drugs we take post-transplant it can be the most problematic. More times than not despite the symptoms it is a necessary evil. Tac / Prograf is very important in helping to adjust your immune systems so you don’t reject your new organ. If your symptoms match some of the symptoms listed above you are probably about average. However, call your doctor right away if you notice any of the following side effects: Allergic reaction: Itching or hives, swelling in your face or hands, swelling or tingling in your mouth or throat, chest tightness, trouble breathing. Change in how much or how often you urinate, painful urination, bloody urine. Confusion, weakness, uneven heartbeat, trouble breathing, numbness in your hands, feet, or lips. Dizziness, lightheaded, fainting. Fast, slow, or pounding heartbeat. Fever, chills, cough, stuffy or runny nose, sore throat, body aches. Headache, vision changes, tremors, seizures, tingling or numbness. Increased thirst or hunger. Rapid weight gain, swelling in your hands, ankles, or feet. Skin changes or growths. Swollen glands in your armpits or groin. Unusual bleeding, bruising, or weakness. Weakness on one side, confusion, clumsiness, trouble thinking clearly.
