What is ONDANSETRON (Zofran)? What is Ondansetron used for? Uses and Side Effects!

Ondansetron, also known as Zofran®, is a medication primarily used to prevent and treat nausea and vomiting resulting from chemotherapy, radiation therapy, or surgery. It comes in several oral forms:

Tablets of 4 mg and 8 mg
A clear liquid solution
A white, round, disintegrating tablet known as ondansetron ODT, in 4 mg and 8 mg doses, which dissolves on the tongue
Additionally, ondansetron can be administered as a clear liquid via intravenous (IV) infusion.

Potential Side Effects of Ondansetron

Common side effects include:

Headaches
Abdominal pain
Diarrhea or constipation
Fatigue
Blurred vision
Skin rashes
Dizziness
Temporary liver function changes
These are not all the possible side effects, and any side effects should be reported to a healthcare provider.

Specific Instructions for Ondansetron Use

If ondansetron causes headaches, acetaminophen (Tylenol®) may be recommended for relief.
For the orally disintegrating tablet (ODT), do not push the tablet through the foil. Instead, peel the foil with dry hands and gently remove the tablet, placing it on your tongue to dissolve quickly. No liquid is needed for this form.
Ondansetron can control nausea and vomiting for up to 8 hours. If you experience nausea or vomiting between doses (known as “breakthrough” symptoms), inform your healthcare provider for potential additional medication.
Store ondansetron tablets and liquid at room temperature in a dry area.

Laboratory Testing and Ondansetron

The body's enzyme, cytochrome P450 2D6 (CYP2D6), is capable of metabolizing various medicines, including ondansetron. Genetic testing can reveal if your CYP2D6 metabolizes medicines at an abnormal rate. If your body processes medicines faster than usual, you might be advised against using ondansetron, as it may not be effective. Consult your healthcare provider for more information on CYP2D6 and its impact on ondansetron.

▪ Antiemetics are a group of medications that are used to treat nausea and vomiting. These include ondansetron, metoclopramide, aprepitant, prochlorperazine, and promethazine, as well as antihistamines like diphenhydramine, and cannabinoids, such as dronabinol.
▪ Ondansetron (Zofran), which can be administered orally, intravenously, and intramuscularly. Once administered, ondansetron acts as a 5-HT3 receptor antagonist peripherally by decreasing vagal nerve stimulation, but it is also a very powerful central acting antiemetic blocking an area in the brain called chemoreceptor trigger zone, or CTZ.
▪ Ondansetron is primarily used to control nausea and vomiting after surgical anesthesia, as well in individuals undergoing chemotherapy or radiotherapy.
▪ . Ondansetron can cause headaches, gastrointestinal disturbances like constipation, and can prolong the QT interval, leading to arrhythmias.
A life-threatening side effect of ondansetron is serotonin syndrome, which is caused by serotonin accumulation that results in overstimulation of the nervous system.
▪ As far as contraindications go, ondansetron should be avoided in clients with risk factors for QT prolongation or in combination with other medications that increase serotonin levels, and should be used with caution in clients with electrolyte abnormalities.

What is Ondansetron? Ondansetron, sold under the brand name Zofran among others, is a medication used to prevent nausea and vomiting caused by cancer chemotherapy, radiation therapy, or surgery. It is also effective for treating gastroenteritis. It can be given by mouth or by injection into a muscle or into a vein.
Side Effects of Ondansetron, Common side effects include diarrhea, constipation, headache, sleepiness, and itchiness. Serious side effects include QT prolongation and severe allergic reaction. It appears to be safe during pregnancy but has not been well studied in this group. It is a serotonin 5-HT3 receptor antagonist. It does not have any effect on dopamine receptors or muscarinic receptors.
Ondansetron was patented in 1984 and approved for medical use in 1990.[8] It is on the World Health Organization's List of Essential Medicines.[9] It is available as a generic medication.[4] In 2019, it was the 60th most commonly prescribed medication in the United States, with more than 11 million prescriptions.

Experienced personal injury attorney Marc Whitehead discusses the drug Zofran, what makes this medication so controversial, and what your options are if you or your child have experienced harmful side effects due to its usage.

Whats Zofran?- The multi- million dollar pharmaceutical giant glaxosmithkline developed Zofran nearly 25 years ago to help cancer patients manage extreme nausea and vomiting in 2006 the US food and Drug administration approved generic drugs to Zofran. For reasons that are debated, Doctors began prescribing the drug for off label uses. "Off label" refers to the use of the drug for something other then what the drug was officially approved to treat.

The most prominent of these uses involved treating pregnant women who had extreme morning sickness. This extreme morning sickness typically presence early in the pregnancy and happens in about 1 of every 100 pregnancies and can cause severe risks to both the mother and the fetus. Doctors thought that Xofran could be useful to treat morning sickness for several reasons.
1- At the time physicians didn't have a go to drug to help pregnant women with this problem.
2- Xofran is technically known as a 5HT3 antagonist meaning it suppresses the release of an important neurotransmitter caller serotonin and at the same time forces the stomach to empty faster thereby reducing the likelihood of painful vomiting.
3- glaxosmithkline educated doctors to view the drug as a high effective and appropriate remedy to stop nauseous behavior in pregnant women.

What went wrong? There have been plenty of ugly facts that have emerged over time to challenge this particular pharmaceutical avenue for treating morning sickness.
1- The FDA never approve Xofran to be used by pregnant women
2- The safety data of this drug was pooled from just over 200 fetal records- a small sample size.
Think about that for a second- million of pregnant women and their fetuses have been subjected to this drug yet the safety data com. es from just over 200 fetuses. That is, by any measure, a staggeringly thin body of evidence.

Ondansetron, marketed under brand names such as Zofran, Zofran ODT, and Zuplenz, is a medication used to prevent nausea and vomiting associated with cancer chemotherapy, radiation therapy, and surgery. It functions as a serotonin 5-HT3 receptor antagonist, blocking serotonin's action in the chemoreceptor trigger zone and on vagal nerve terminals in the gastrointestinal tract. Common side effects include headache, dizziness, drowsiness, constipation, and fatigue, while serious side effects may include QT prolongation, serotonin syndrome, and allergic reactions. Dosages vary based on the condition: for chemotherapy-induced nausea, adults typically take 8 mg twice daily or 24 mg as a single dose, and for radiation-induced nausea, 8 mg orally before radiotherapy.
