Successful drug for multi-drug resistant strains of gram-positive bacteria like MRSA, VRSA. Works on 50S subunit in such a way that it can't bind with 30S subunit, and functional initiation complex can't be formed.

Aminoglycosides are antibiotics used for gram negative infections. Gram-negative means that the bacteria which cause the infection don't hold the colour of a particular dye (Crystal Violet) when they're tested.

Since they are not absorbed from the gut, they are usually taken by an injection into the vain or muscle but they can be given as a cream or ointment to prevent and treat skin infections, as eye drops for eye infections, as ear drops for ear infections and as a pill for intestinal infections but not for other internal infections.

Antibiotics from this family can be used in patients who are not able to take some regular antibiotics, such as Penicillin because of allergies. The most common Aminoglycosides used as alternatives to Penicillin are Erythromycin (lit. Red fungus) and Clarithromycin (lit. Yellow fungus). They're most commonly used in the treatment of bacterial infections, such as Bronchitis.

Linezolid is an antibiotic used for the treatment of infections caused by Gram-positive bacteria that are resistant to other antibiotics. Linezolid is active against most Gram-positive bacteria that cause disease, including streptococci, vancomycin-resistant enterococci (VRE), and methicillin-resistant Staphylococcus aureus (MRSA). The main uses are infections of the skin and pneumonia although it may be used for a variety of other infections including drug-resistant tuberculosis. It is used either by injection into a vein or by mouth.

When given for short periods, linezolid is a relatively safe antibiotic. It can be used in people of all ages and in people with liver disease or poor kidney function. Common side effects with short-term use include headache, diarrhea, rash, and nausea. Serious side effects may include serotonin syndrome, bone marrow suppression, and high blood lactate levels, particularly when used for more than two weeks. If used for longer periods it may cause nerve damage, including optic nerve damage, which may be irreversible.

As a protein synthesis inhibitor, linezolid works by suppressing bacterial protein production. This either stops growth or results in bacterial death. Although many antibiotics work this way, the exact mechanism of action of linezolid appears to be unique in that it blocks the initiation of protein production, rather than one of the later steps. As of 2014, bacterial resistance to linezolid has remained low. Linezolid is a member of the oxazolidinone class of medications.

Linezolid was discovered in the mid-1990s, and was approved for commercial use in 2000.

She started feeling fullness/pressure in her pelvis and scans eventually revealed a grapefruit-sized mass in her pelvis, with part of it being a solid tumor. After surgery to remove the mass, the pathology revealed her diagnosis as extra ovarian primary peritoneal carcinoma, which is a rare gynecologic cancer.

Amanda is in full remission, but she will continue on the immunotherapy for another year.

This drug Hydroxyurea – that was really the basis of our work in the late 80’s and early 90’s.



There was a group in the Heart, Lung, and Blood Institute who started working on this using a drug called 5-Azacytidine.


Just like all chemotherapy, it first causes some toxicity to the rapidly dividing cells, and then when you withdraw the drug and you give the cells a chance to recover maybe that’s what’s turning on the genes. And that led us to actually consider this drug Hydroxyurea because it’s a form of chemotherapy. It works on rapidly dividing cells. It doesn’t have the same toxicity profile that drugs like 5-Azacytidine does.

Using this drug Hydroxyurea in about 70 percent of hospitalized patients we could turn on their fetal hemoglobin to modest levels and the longer you gave it to them, the higher their values would go. Subsequently our work was replicated by groups in Seattle and Harvard and Johns Hopkins, and that led the NIH to fund this fairly large study, a cooperative randomized clinical trial, which tests whether the drug would in fact have a benefit.

And just to cut to the chase, the study was stopped early, a year early, because one group, which was blinded at the time, was doing much much, better than the other group, and that group was the Hydroxyurea group. In 1990, our paper came out. In 1995, this NIH-supported trial, this large randomized trial, showed that Hydroxyurea was quite beneficial. And in 1998, the FDA approved the drug as the first drug that was approved specifically to treat patients with sickle cell disease.

Long off patent, glipizide is an inexpensive sulfonylurea drug approved by the FDA in 1984. With more than 17 million prescriptions each year in the United States, glipizide ranks among the 50 most frequently dispensed drugs. Due to glipizide's low price, pharmaceutical companies reserve their advertising dollars for more profitable products. As a result many doctors and patients alike simply overlook this reasonable option.

Most of the anti-diabetic drugs including glipizide reduce the glycosylated hemoglobin or A1c to a similar degree. Metformin generally should be the first therapeutic option beyond diet and exercise with many individuals ultimately requiring a second and even third drug. Glipizide may be purchased either separately or in combination with metformin.

Side effects associated with glipizide remain relatively few and easily managed. Glipizide appears less likely to result in hypoglycemia than other drugs in the same family. Glipizide should not be rank as a primary agent for consideration among those with pre-existing heart disease.
