Pioglitazone, sold under the brand name Actos among others, is an anti-diabetic medication used to treat type 2 diabetes.[2] It may be used with metformin, a sulfonylurea, or insulin.[2][3] Use is recommended together with exercise and diet.[3] It is not recommended in type 1 diabetes.[3] It is taken by mouth.[3]

Common side effects include headaches, muscle pains, inflammation of the throat, and swelling.[3] Serious side effects may include bladder cancer, low blood sugar, heart failure, and osteoporosis.[3][2] Use is not recommended in pregnancy or breastfeeding.[2] It is in the thiazolidinedione (TZD) class and works by improving sensitivity of tissues to insulin.[2]

Pioglitazone was patented in 1985, and came into medical use in 1999.[4] It is available as a generic medication.[2] In 2019, it was the 179th most commonly prescribed medication in the United States, with more than 3 million prescriptions.[5][6] It was withdrawn in France and Germany in 2011.[7][8][9] Pioglitazone is used to lower blood glucose levels in type 2 diabetes either alone or in combination with a sulfonylurea, metformin, or insulin.[1] The effects of Plioglitazone have been compared in a Cochrane systematic review to that of other blood sugar lowering-medicine, including metformin, acarbose, and repaglinide, as well as with appropriate diet and exercise, not showing any benefit in reducing the chance of developing type 2 diabetes in people at risk.[10] It did, however, show reduction of risk of developing type 2 diabetes when compared to a placebo or to no treatment.[10] These results should be interpreted considering that most of the data of the studies included in this review were of low or very-low certainty.

While pioglitazone does decrease blood sugar levels, the main study that looked at the medication found no difference in the main cardiovascular outcomes that were looked at.[11] The secondary outcome of death from all causes, myocardial infarction, and stroke were lower.[11]

Pioglitazone has been found to reduce all-cause mortality in type 2 diabetic patients compared to other therapies, with a 60% reduction in mortality in those exposed to pioglitazone, compared to those never exposed.[12] Another study found an all-cause mortality hazard ratio of 0.33 for pioglitazone after adjusting for 40 covariates, compared to insulin.[13] Due to insufficient data on all-cause mortality, cardiovascular mortality, myocardial infarction and stroke, this was not possible to compare in a more recent review.[10]

Phenytoin (Dilantin) – Side Effects, Uses, Mechanism of Action, Dosage, Interactions, Warnings
Phenytoin (Dilantin) is an anticonvulsant medication used to control seizures, particularly tonic-clonic (grand mal) and complex partial seizures, as well as seizures following neurosurgery. It works by stabilizing neuronal membranes and inhibiting sodium channels, reducing seizure activity in the brain. Common side effects include dizziness, nausea, and gum overgrowth, while serious effects can include severe skin reactions, blood disorders, and liver toxicity. The dosage varies, typically 100-400 mg daily for adults, with therapeutic monitoring required due to its narrow therapeutic range. Phenytoin interacts with many drugs, including contraceptives and warfarin, and poses risks such as cardiovascular issues, teratogenicity, and bone density loss, especially with long-term use. Careful consideration is needed for patients with liver impairment and during pregnancy.

Phenytoin - Uses, Mechanism Of Action, Adverse Effects & Toxicity (Anti-epileptic Drugs)
Phenytoin - Uses, Mechanism Of Action, Adverse Effects & Toxicity (Anti-epileptic Drugs) Side Effects 1 Views • 2 months ago

Phenytoin is a first-generation anti-epileptic medication used in the treatment of various types of seizures, terminal neuralgia, behavioral disorders, and status epilepticus; a condition characterized by prolonged seizures.
A seizure is a condition where there is uncontrolled, abnormal electrical activity of the brain, causing neurons to fire action potentials more frequently.
Voltage-gated membrane sodium channels of the neurons are responsible for this increased firing of nerve signals.
Phenytoin works by blocking these voltage-gated sodium channels, reducing the frequency of nerve signals.
Phenytoin can be administered orally and intravenously.
Phenytoin achieves higher concentrations in the central nervous system compared to the serum.
Therefore, it is ideal in the treatment of central nervous system disorders.
Phenytoin is metabolized by the cytochrome P450 enzyme system in the liver.
Drugs that alter the function of these enzymes affect the plasma concentrations of phenytoin.
Medications that inhibit these enzymes will increase the plasma concentrations of phenytoin.
Some of these drugs include amiodarone, cimetidine, metronidazole, chloramphenicol, sodium valproate, and sulfonamides.
By contrast, medications that induce the enzyme system will decrease the plasma concentrations of phenytoin.
Some of these drugs include rifampicin, barbiturates, theophylline, and carbamazepine.
So, individuals who are taking these medications concurrently with phenytoin require dose adjustments according to the clinical response.
Major adverse effects of phenytoin include the following.
Gingival hyperplasia.
Hypotension.
Arrhythmias.
Sedation.
Psychosis.
Megaloblastic anemia.
Steven Johnson syndrome.
Decreased bone mineral content with long-term use.
And immunoglobulin A deficiency.
Since phenytoin is a medication with a narrow therapeutic index, it is important to know the toxic effects of the drug.
Phenytoin displays its primary signs of toxicity in the nervous system and cardiovascular system.
Main central nervous system effects include nystagmus, slurred speech, tremors, hyperactivity, seizures, and coma.
In the cardiovascular system, it can cause life-threatening arrhythmias, hypotension, and cardiovascular collapse.
Finally, there is a rare condition associated with phenytoin crystallization within the circulation called “Purple Glove Syndrome”.
This condition is characterized by worsening limb edema and discoloration.
If not identified and corrected early, this can lead to life-threatening ischemia of the limb, which necessitates amputation.

Today we're going to learn about Samantha and her weight loss journey on Phentermine.

Phentermine has been shown to be effective in promoting weight loss in overweight and obese individuals, it is important to note that a healthy diet and exercise routine are crucial components in achieving long-term weight loss and overall health.

Phentermine has been used by many people, including celebrities, to help them lose weight and achieve their weight loss goals. However, it should be noted that phentermine is not a magic pill and should always be used in conjunction with a healthy diet and exercise routine. There have been numerous scientific studies that have proven the effectiveness of phentermine for weight loss, but it should only be used for a limited period of time and under the supervision of a doctor. Maintaining a healthy lifestyle, including a balanced diet, regular exercise, and healthy habits, is crucial for long-term weight loss and overall health.

Phentermine or Adipex P Medication Information (dosing, side effects, patient counseling)
Phentermine or Adipex P Medication Information (dosing, side effects, patient counseling) Side Effects 1 Views • 2 months ago

Your Medication Phentermine

Phentermine is also known by the brand name: Adipex P.

Phentermine comes in 15, 30, 37.5 mg capsules, and 37.5 mg tablets and should be stored at room temperature in a dry place.

Phentermine is most commonly used to treat obesity. No matter what your doctor has you taking it for, phentermine is taken by mouth once daily. Take all forms before breakfast or 1 to 2 hours after breakfast. If you miss a dose, take the missed dose as soon as you think about it. If it is close to time for your next dose, skip the missed dose and resume your normal dosing schedule. Do not take 2 doses at the same time or extra doses.

Some common side effects with phentermine include high blood pressure, headache, dizziness, restlessness, and increased heart rate.
Some less common side effects include constipation, unpleasant taste, dry mouth and unable to get to sleep at night.

Do not take phentermine if you have a history of cardiovascular disease, hyperthyroidism, glaucoma, a history of drug abuse, or are pregnant or breastfeeding.
While taking phentermine, remember it is important to tell your doctor or pharmacist if you:
• Have signs of an allergic reaction (rash, itching, swelling, tightness of the chest, etc.)
• Experience behavioral changes
• Experience chest pain, increased heart rate, tremors
• Have severe headache
• Have swelling of the arms or legs
• Start taking new medications, vitamins, or supplements

Phentermine is an FDA approved oral therapy that assists in short-term weight loss in combination with diet and exercise. Phentermine belongs to the appetite suppressing family and is a weak relative of amphetamine. Phentermine may be appropriate when the BMI – body mass index – exceeds 30 or 27 in the presence of other cardiovascular risk factors .

Phentermine is inappropriate for those who desire to lose a few pounds. In selected individuals it should be administered on hour or two before or after breakfast. A split dose of 18.75 mg before breakfast and dinner is also acceptable. Since phentermine functions as a stimulant, a bedtime dose risks insomnia.

Therapy may continue for a maximum of 12 weeks. Unfortunately a regain of weight generally occurs in the absence of a continuous low calorie diet. weight regain occurs after discontinuation of therapy. The greatest weight loss occurs in the first several weeks of therapy. Effectiveness of Phentermine is lost beyond 12 weeks of use.

Contraindications against use of phentermine include coronary artery disease, arrhythmia, uncontrolled blood pressure and heart failure. Hyperthyroidism, glaucoma, an agitated state and pregnancy also exclude Phentermine eligibility. Phentermine should not be prescribed to those with a history of drug abuse.

Generally well tolerated, adverse effects of phentermine include dry mouth, unpleasant taste, diarrhea or constipation. Hives or urticaria, hair loss and erectile dysfunction may also develop. Symptoms of acute phentermine overdose present as restlessness, tremor, panic attacks, confusion and hallucinations. Insomnia, irritability, hyperactivity and psychosis reminiscent of schizophrenia may indicate chronic toxicity.

STOP Taking Phentermine (It Doesn't Work)
STOP Taking Phentermine (It Doesn't Work) Side Effects 1 Views • 2 months ago

3 reasons why Phentermine is harmful to your body. This list of phentermine symptoms may cause you to think twice before using this weight loss medication.

Phentermine is one of the most common weight loss medications prescribed by doctors. But there's one big problem:

It doesn't work nearly as well as you might think.

Yes, it can help you lose weight but that weight loss is only temporary. In addition, it comes at the cost of damage to your metabolism, a loss in lean muscle mass, and it may also cause some serious side effects.

Before you use phentermine for weight loss, you need to watch this video so you can avoid these negative side effects.

There are other weight loss medications available which can help with long-term weight loss which do NOT cause these negative side effects.

The Most Common Mistake Patients Make with Phentermine Side Effects 1 Views • 2 months ago

Dr Covington discusses the most common mistake that patients make with phentermine and why avoiding this mistake will improve not only short term but also long term weight loss success.

Failing to eat or skipping multiple meals throughout the day is a bad idea with phentermine, it is perhaps one of the worst things that you can do, here's just a few reasons why:

The 5 reasons Dr Covington gives for why skipping meals when taking phentermine is bad:

1. Leads to Poor Nutrition and Potential Nutrition Deficiencies

2. Increases Cravings

3. Increases Hunger

4. Leads to Loss of Muscle and Slow Metabolism

5. Creates a Failure to focus on long term weight loss success as opposed to short term weight loss

CBD Oil and Seizures in Dogs (the best treatment?) - Dog Health Vet Advice
CBD Oil and Seizures in Dogs (the best treatment?) - Dog Health Vet Advice Side Effects 1 Views • 2 months ago

Can you use CBD oil to treat seizures and epilepsy in dogs? If so how do you give it and what dose of CBD oil should you use?

Here are some high-quality CBD options with independent analysis:
- Oil:
https://cbdmd.t7c9v8.net/XD25X
- Hard chews: https://cbdmd.t7c9v8.net/0LRBR
- Soft chews: https://cbdmd.t7c9v8.net/EoRY2
- Peanut butter: https://cbdmd.t7c9v8.net/6rV9E

- Need to try and determine the causes of the seizures - organ disease, infections, poisoning, brain lesions, epilepsy

- When to start treatment if epilepsy - 2 or more seizures in 6 months - certainly more than 1 a month, not recovering between seizures, severe seizure or prolonged recovery
If not epilepsy then need to try and address underlying cause as often don’t respond as well to anti-seizure medication alone

- Need to pick best anti-seizure medication
With some drugs like phenobarbital and imepitoin we know how effective they can be and what to expect with long term administration. With CBD oil research into its use to control seizures is in the early stages, there are currently no published studies detailing how effective it is, although these are starting to be run.

- Why try a drug we don’t know much about or how well it works over other drugs that we know work in a large number of patients?

Nurokinin 1 NK-1,
Cannabinoids CB-1,
serotonin 5-HT3,
dopamine D2,
muscarinic M1
and histamine H1 receptors.
The vomiting center can be activated by afferent fibers
arising from the gastrointestinal track,
Chemoreceptor trigger zone CTZ,
Cerebral cortex and vestibular apparatus.
The CTZ or Chemo receptor trigger zone, is located at the end of fourth ventricle.
CTZ have receptor of serotonin 5-HT3, dopamine D2, muscarinic M1 receptors.
It lies outside of blood brain barrier and stimulate vomiting center in response to
blood borne substances including cytotoxic cancer chemotherapy drugs.
The excessive vestibular stimulation in motion sickness activates the vomiting center
via cholinergic and histaminergic nerve fibers that projected through vestibulocerebellar region of cerebellum.
The irritation of gastric mucosa by chemotheraperautic agents, endogenous toxin and poison leads to
secretion of 5-HT which activate 5-HT3 receptor and this stimuli travel from the gastrointestinal tract

Antihistaminic
Antihistaminic antiemetic includes
Cinnarizine, cyclizine and promethazine
Antihistaminic drugs blocks muscarinic M1 receptors or histamine H1 receptors or both types of receptors
in the vestibular tracts that project to the vomiting center and in the CTZ.
Antihistaminic drug Cyclizine is used in motion sickness.
cinnarizine is used in motion sickness and vestibular disorders such as Menière’s disease)
promethazine is used in severe morning sickness of pregnancy.

Antimuscarinics
Antimuscarinics drug Scopolamine is used in motion sickness.
It causes blocking of muscarinic M1 in vestibular tracts.

5-HT3 receptors antagonist
5-HT3 receptors antagonist includes Granisetron, ondansetron, dolasetron, tropisetron and palonsetron.
They are particular value in preventing and treating the vomiting and nausea.
commonly encountered postoperatively as well as that caused by radiation therapy or administration of cytotoxic drugs such as cisplatin.
They prevent emesis by competitively blocking 5-HT3
